to achieve best-in-class efficacy and significantly reduce the dosing burden for patients across the largest inflammatory and

immunology markets.

Advanced Antibody Engineering: The FcRn Recycling Platform

Apogee's clinical-stage pipeline is built on a proprietary antibody engineering platform designed to drastically reduce the

patient injection burden in immunology and inflammation (I&I) markets. The foundational technology focuses on supercharging

the human body’s natural antibody recycling mechanism through the neonatal Fc receptor (FcRn). Normally, antibodies in

circulation are internalized by cells and degraded, which limits their therapeutic half-life. To counter this, Apogee systematically

evaluates and introduces two distinct half-life extension technologies into the Fc region of its monoclonal IgG1 antibodies:

• YTE Mutation Technology: This approach introduces a specific triple amino acid modification (M252Y/S254T/T256E) into

the antibody's Fc domain. By enhancing binding affinity to FcRn at acidic pH within endosomes, YTE-modified antibodies

are spared from intracellular lysosomal degradation and are recycled back into active circulation. Furthermore, third-party

clinical trials indicate that the half-life extension conferred by YTE is highly consistent from patient to patient, reducing

pharmacokinetic (PK) exposure variability.
