On the Launch Radar: Physician Insights on IONS’ Tryngolza sHTG- quotes & transcript

sHTG market opportunity? Well, I thought I’d just outline some of the quick debates that

we have with investors regarding this category build and opportunity.

The bull case is that sHTG can be a large market, aka TAM, with a big unmet need for

better TG, or triglyceride lowering therapies, that the APO-C3 class has shown an

unprecedented TG lowering, and that Tryngolza is the first drug to show a reduction in

acute pancreatitis from an event rate perspective. And so these things have afforded the

company an ability to come to the market with a premium price of $40,000 per annum

on a list price basis.

The bear case arguments are going to be, or that we hear at least, is that sHTG is a

silent disease. What is patient adherence and persistence going to be like to a new

monthly therapy? Could the launch be slow? What are the kinetics going to look like for

patients accessing new medicines, again, given this is a silent disease in most

instances? What percentage of patients are going to need a new APO-C3 drug? There

are some safety debates around liver fat. Is that a signal? Is it transient?

And then there's obviously going to be debates about Arrowhead’s Redemplo, which has

less frequent dosing. It, at least at its Phase 3 study dose, doesn't appear to have a liver

fat signal. So certainly that's something that investors will debate ahead of Arrowhead’s

Phase 3 SHASTA-3 and 4 results in the third quarter of this year.

Quickly, this slide here, there is certainly no shortage of debates on how big of a market

really is this. A lot of the epidemiology cuts and segmentation have been given to us by

Ionis, and so this is just a spectrum of scenarios where the most high-risk patients, who
