geoffrey.vonderahe@wedbush.com 15% of patients and is characterized by the development of subcutaneous
nodules composed of monosodium urate (tophi), which result in inflammation and
potential joint erosion and deformity.
Hyperuricemia is a necessary risk factor for gout, and occurs due to the
overproduction or most commnly underexcretion of uric acid. In addition to
endogenous production and underexcretion, consumption of purine-rich foods,
including alcohol, red meat and seafood, can also promote hyperuricemia.
However, dietary modifications in patients typically result in only modest
reductions in serum urate, and suffer from low rates of compliance. Therapies
used to manage acute gout flares and urate lowering therapies (ULTs) have
contraindications with comorbidities, and gout disproportionately affects older
adults, who frequently have comorbid conditions such as chronic kidney disease
and cardiovascular disease. A subset of patients fails to achieve serum urate target
goals owing to intolerance or inadequate response to available agents.
Here, we provide an overview of gout, the current and emerging treatment
landscape, and highlight companies working in the space. These include
Monte Rosa Therapeutics (GLUE, OUTPERFORM) developing Nek7 molecular glue
degrader MRT-8102 for the treatment of acute gout flares and potentially anti-
