We're kicking off our ADA conference coverage with the dose finding angle of GPCR's ACCESS trial and impact of
NOVOB's CagriSema on patients appetite and brain activity. On the latter, we learned valuable insights applicable

Aleniglipron's dose-finding results provide valuable insights for the planned phase 3. Recall, Structure (GPCR)
is evaluating aleniglipron, a small molecule GLP-1, for obesity and diabetes. In the phase 2b ACCESS trial, GPCR
was investigating the efficacy and safety in patients with obese and/or overweight and ≥1 weight-related comorbidity
(n=230) for 36 weeks, with the primary endpoint of change in body weight from baseline (Abstract 1032-OR). To set
the stage, in the trial, there was mean BMI of 39.5 kg/m2, and 54.3% of participants were female.
●Efficacy: At week 36, at doses of 45, 90, and 120 mg, aleniglipron induced absolute weight loss from baseline
of -9.0%, -10.7%, and -12.1%, respectively, and -0.8% for placebo. Per the investigator, there were no signs of
plateauing, and in the ongoing open label extension, there was suggestive evidence of additional weight loss of
up to 16.2% in the interim analysis after a median of 20 weeks. Importantly, 86% of patients lost ≥5% of their
weight, 70% lost ≥10%, and 38% lost ≥15% in the 120 mg dose arm. To provide some context, this weight loss
was relatively comparable to Lilly's (LLY) Foundayo (orforglipron), cross trial comparison caveats aside, which in
