To support NCCN inclusion the company is set to publish an initial

manuscript of SENTRY results in a peer-reviewed journal in the upcoming

months (mid-2026) and will be presenting fuller SENTRY data at ASCO.

We believe the ASCO messaging will focus on further connecting the

dots between spleen volume reduction, variant allele frequency (VAF)

reductions, and overall survival. Additionally, we think there is the potential

for other post-hoc analyses such as exploring the impacts of total symptom

score (TSS) baselines on absolute improvement in symptoms given that this

was a co-primary that missed statistical significance.

Overall, based on additional commercial work we conducted (note here),

we do believe that NCCN inclusion would likely lead to meaningful use of

selinexor; though from a commercial modelling perspective we remain on

the sidelines until we see initial sales ramp up to better inform forecasts.

Our prediction for XPORT-EC-042 top line: per our proprietary modeling

presented in our prior initial XPORT note, we predict with an 80%+ PoS that

the study will show at least an HR of 0.5-0.6, which well exceeds physician

expectations and hurdle for adoption.

Furthermore, on today’s call management provided additional color that

median follow-up time for the study should likely fall between SIENDO

(prior EC maintenance study) top line and the initial longer-term data

cut follow-up times, which ranged from ~10-29 months; the mid-point of

this falls neatly in our median follow-up time estimates, supporting our

modeling assumptions and our confidence in our initial predictions.
