Bringing NK Cell Therapy To Autoimmune Disease

Autoimmune disease is increasingly emerging as a compelling next frontier for cell

therapy, driven by the growing recognition that many severe, refractory indications

are fundamentally mediated by pathogenic B cells and remain inadequately

addressed by current standards of care. Despite affecting ~7 million patients in the

U.S., treatment today is largely centered around chronic, outpatient

immunosuppression that rarely induces durable remission and is often constrained

by cumulative toxicity. Encouragingly, recent results with CD19-directed cell

therapies from academia and companies have begun to challenge this paradigm,

showing that a one-time B-cell-depleting intervention can drive deep, drug-free

remissions lasting multiple years across diseases including lupus, myositis and

systemic sclerosis. Mechanistically, transient depletion of autoreactive B cells

followed by reconstitution with naïve populations may enable a functional "immune

reset," offering the potential to alter disease course rather than simply manage

symptoms. Taken together, these data have catalyzed significant interest in applying

cell therapy to autoimmune disease and support a shift toward potentially curative,

one-time treatment approaches.
