500 - First results from the OPTIMA phase III randomized non-inferiority trial of test-

directed chemotherapy in patients with high clinical risk ER-positive HER2-negative

early breast cancer

This is the pivotal trial for VCYT's Prosigna LDT. Pending a positive trial • Context:

readout, VCYT would launch the test mid-year 2026 and increase commercial headcount

to support the assay based on the strength of the data. VCYT has previously outlined

the goals of OPTIMA as being to (1) replicate the predictive claim of Oncotype Dx as

established in TAILORx, and (2) expand on the intended use population in terms of nodal

status and inclusion of premenopausal and postmenopausal patients.
