A few interesting news items in obesity space this week. Small-molecule Ph2
China data showed ~11% WL at 36W with decent tolerability; VKTX initiated
VK3019 (DACRA) in HV backed by impressive preclinical result; WVE INHBE
initiated Ph2a multidose portion of INLIGHT. Ascletis' ASC35 - monthly GLP-1/
GIP IND cleared to start Ph1 SAD/ MAD which include H2H design against tirze.
LLY provides more clarity on Medicare GLP-1 Bridge program to take effect July
1st, '26.
Bottom line. This week’s obesity news flow highlights four key strategic directions shaping the
market: oral incretins, amylin/ DACRA, novel MOAs, and long-acting.
For oral incretin, we think the recent progress - particularly two oral launch - further validate
the feasibility of this category. However, emerging late-stage clinical data suggest differentiation
increasingly occurs between programs within the category rather than between modalities. Clear
leaders/ new benchmarks have emerged - namely GPCR's aleni for small molecule, with ~15% at
36W (HD) reaching injectable levels, while others risk converging toward a “good enough” profile.
Recent small molecule China data showing ~11% weight loss at 36W are broadly in line with prior
oral GLP-1 benchmarks, with good tolerability and low d/c. That said, aleni wins efficacy, and has
a large therapeutic window without scaffold-related safety overhang. Overall, we see aleni remains
the leader on efficacy with GI toleratbility effectively addressed by milder dosing regimen, potentially
meaningful share vs. later "viable" entrants.
