acute bipolar mania.BIOTECHNOLOGY WHAT YOU SHOULD KNOW: Despite decades of therapeutic innovation, treatment
of acute bipolar mania remains largely dependent on dopamine-modulating
antipsychotics and mood stabilizers developed between 1970 and 2015. While modern
agents have improved tolerability and expanded treatment across multiple phases of
bipolar disorder, meaningful challenges remain including akathisia, metabolic burden,
sedation, orthostatic hypotension and medication adherence. This vacuum in novelINDUSTRY mechanism driven assets continues to be a meaningful commercial opportunity for
agents that can demonstrate robust antimanic efficacy while offering differentiated
safety and tolerability advantages. RAP-219’s TARPγ8-selective mechanism is well-
matched to the neurobiology of acute mania and its selectivity could proveREPORT advantageous to mitigating these unwanted side effects.
Acute mania is one of the most disruptive manifestations of bipolar disorder and
frequently results in psychiatric hospitalization, increased healthcare utilization,
treatment non-compliance and elevated risk of dangerous behavior or legal
involvement.
■ Current treatment of acute bipolar mania revolves around mood stabilizers and
atypical antipsychotics that have been SOC for the past two decades. Most clinical
practice guidelines recommend lithium or divalproex as foundational therapies,
with atypical antipsychotics added in moderate-to-severe episodes or when rapid
