We hosted an investor call with Dr. John Kaczmar, M.D., Medical Oncologist and
Associate Professor at the Medical University of South Carolina, specializing in treating
cancers of the head and neck.
Our KOL, Dr. Kaczmar, noted that he currently treats a few dozen 1L R/M HNSCC
patients per year and that a majority of his patients have HPV-negative disease. Given
Dr. Kaczmar's practice is located in the south-east portion of the US (South Carolina), rates
of smoking are higher than in other regions of the US, so a majority of his patients have HPV-
negative disease with a smaller subset being the healthier HPV-positive subset. Regarding
the breakdown of CPS status in his 1L R/M population, he notes that approximately 5%
of his patients are CPS-negative, similar to rates seen in KN-689, which differs from other
reported rates in the literature which are closer to 15%. As such, Dr. Kaczmar suspects the
vast majority of patients are CPS-positive. If a patient is not placed on a clinical trial, the
commercial options Dr. Kaczmar opts for differs based on a patients CPS status, namely
varying based on CPS-high or -low delineation. For the 5% of patient he sees that are CPS-
negative, Dr. Kaczmar elects to use Keytruda pembrolizumab [MRK] with chemo rather
than cetuximab based on his own experiences where CPS test results may have varying
