New Large-Scale Study Further Exonerates Tylenol in Pregnancy: A landmark population-based cohort study published in JAMA

Internal Medicine (published 6/29/26) adds significant weight to the growing body of evidence that prenatal acetaminophen

(Tylenol) use does not cause autism spectrum disorder (ASD) or attention deficit-hyperactivity disorder (ADHD) in children.

Conducted by Eric Yuk Fai Wan, PhD, and colleagues at the University of Hong Kong, the study analyzed electronic health records

from over 708,000 mother-child pairs between 2001 and 2023 — one of the largest and most methodologically rigorous studies

on this topic to date.

Key Findings: The headline result from sibling-matched analyses — the gold standard design for controlling familial confounders

— showed no association between prenatal acetaminophen exposure and either autism (adjusted HR 1.00, 95% CI 0.91–1.11)

or ADHD (adjusted HR 1.01, 95% CI 0.93–1.08). These null findings were consistent regardless of timing of exposure during

pregnancy, cumulative dose, or frequency of use. Critically, when the researchers applied a negative control analysis (examining

acetaminophen use in the year before pregnancy), they observed similarly modest positive associations for both autism (HR
