●While some stroke patients can spontaneously improve (e.g.

TIA = transient ischemic attack), this tends to be very uncommon

with larger blockages. Inclusion/exclusion criteria and advanced

diagnostic techniques can be utilized to minimize TIA or other

"stroke-mimic" conditions such as seizures in stroke trials - an

important consideration for being able to detect signal vs noise.

●Morbidity and mortality remain high in untreated AIS patients. Those

who do not receive acute therapies are often faced with significant

disabilities, which can lead to fatal complications, such as pulmonary

embolism and pneumonia.

●In those that are eligible for acute therapy, "time is brain" (meaning

patients need to be treated ASAP). If recanalization is successful,

physicians frequently observe dramatic improvements shortly after

intervention, reinforcing the need for early action.

●For example, patients who were previously unable to speak or

move often can regain these abilities within minutes to hours

after successful restoration of blood flow.

●This is a large market. Existing fibrinolytics are multi-billion dollar

drugs despite reaching fewer than 10% of AIS patients. Basking believes

they could address a substantially larger market with their approach.

Basking's BB-031 is a novel approach to treating AIS:

●BB-031 is an RNA aptamer that selectively binds to von Willebrand

Factor (vWF), blocking its interaction with platelets. vWF functions

to anchor clots to vessel walls, reinforce clot structure, and promote

inflammatory signaling.

●By inhibiting vWF, BB-031 rapidly destabilizes the clot and allows

vessels to reopen. Preclinical studies have demonstrated powerful clot
