platform and take a deeper look at its lead asset, SYX-5219, an oral PKM2

modulator currently in a Phase 1b trial in atopic dermatitis (AD), with a

readout expected in 4Q26.

In healthy volunteers, SYX-5219 has already demonstrated good TARC

(key PD marker in AD) reduction, comparable to IL-4/IL-13 drugs and

Kymera’s STAT6 degrader, giving management confidence in the success

of the upcoming Phase 1b AD trial.

PKM2 could also work in several other indications (e.g., asthma), but we

thought the link between PKM2 biology and obesity-related inflammation

was particularly interesting. Sitryx plans to test this in a basket trial focused

on inflammatory skin diseases.
