The KOL Is Impressed With The AlloNK Data And Believes Efficacy Is Consistent With The

Deep B-Cell Depletion Observed

On the call, the KOL (Dr. Emery) emphasized the significant unmet need in refractory RA,

explaining that as the disease remains uncontrolled over time, persistent inflammation leads

to cumulative joint damage and reduces the effectiveness of subsequent therapies. He

also highlighted his prior 2008 paper evaluating RTX alone, where high-sensitivity assays

demonstrated that B-cell depletion was often incomplete, with response depth correlating with

the speed and magnitude of B-cell depletion.

Against this backdrop, the KOL provided his perspective on the initial Phase IIa dataset, noting

that AlloNK + RTX achieved complete B-cell depletion in all patients (using a high-sensitivity

assay), in contrast to his RTX only data. The treated population was highly severe, with a

median disease duration of ~13 years and elevated baseline CDAI and DAS28 scores (51 and

7.4, respectively) that were significantly above the threshold for high disease activity (>22

and >5.1, respectively). Given this population, he was particularly impressed by the depth of

clinical responses observed to date, as well as the early signs of durability, which he believes

are consistent with the degree of B-cell depletion achieved.
