ASCO 2026: The Only Clones We Want Are Replicate Results From The Phase 2
June 2, 2026
A wall of evidence has supported the development of intismeran across numerous tumor types. The event included
prepared remarks from:
●David Berman, MD, PhD (Chief Development Officer) with an overview of intismeran and why adjuvant therapy
remains a key unmet need across numerous solid tumors
●Michelle Brown, MD (Portfolio Lead, Oncology) on intismeran's mechanism of action and the presence of intended
neoantigens
●Matteo Carlino, MBBS, PhD (presenting author for 5-year KEYNOTE-942 presentation at ASCO) to recap the data
●And Ryan Sullivan, MD (lead author on intismeran poster) showcasing translational data linking the T-cell
clonotypes to encoded neoantigens from intismeran
Translatability from the phase 2 to the phase 3 in adjuvant melanoma trial at the center of investors' minds. In the
phase 2 KEYNOTE-942 trial, we'd remind that 83% of patients on the INT+pembro arm and 84% of patients on the
pembro monotherapy arm were stage IIIC, and there was a far more pronounced benefit among this group versus stage
IV. However, the phase 3 is inclusive of patients across Stage II, III, & IV, and participants were stratified evenly by risk
(as well as age), raising concerns on how replicable the data will pan out to be. Here, it was emphasized by MRNA
that the earlier you go, 1) the more likely you will see a treatment effect and 2) the less events you see. Dr. Carlino
added that his "gut feeling" is the benefit will be the same, with actually some chance that the benefit is actually greater.
Here, he notes that the greater the disease risk, the sooner you get recurrences. Nonetheless, stage IIC should still be
considered a high risk population, per Dr.
