Treatment landscape and importance of QoL in mCRC
●Earlier-line (1L & 2L) mCRC treatment is guided by tumor sidedness and
biomarker selection (BRAF, MSS/MSI, HER2, and RAS status).
●Unfortunately, the late-line (3L+) mCRC treatment setting is unguided
and has few attractive options, with even Lonsurf + bev delivering an
mOS of <1 year.
What are clinically meaningful benchmarks in 3L+ mCRC?
●ORR >5%
●mPFS > 6 months
●mOS >10 months
Quality-of-life matters nearly as much as tumor control:
●Our KOLs were both highly sensitive to safety/tolerability and a
patient's quality-of-life when considering which treatments to use in
the 3L+ setting.
●Double-digit rates of Grade ≥3 adverse events were viewed as difficult
to accept, especially when toxicities lead to ED visits or hospitalization.
