Resend - Modeling CARDIO-TTRansform: Potential Outcome & Implications

properly conducted study to evaluate its drug. PFE is likely to continue
to emphasize its data showing TAF is a strong stabilizer, and as an easy
1/d oral drug with leading market share, suggest it should remain the
preferred treatment option for patients.
Our discussions with TTR-CM-treating cardiologists regarding the
potential CARDIO-TTRansform outcomes and their impact on their
treatment paradigm were frustrating, to say the least.
●They didn't understand the underlying disease biology, the role of
monomers, or the true merits of each therapy.
●They demonstrated indifference to potential longer-term
outcomes in TTR-CM patients since patients are older (which we
found a little offensive) and they blend evidence-based medicine
with non-evidence-based, poorly-informed 'gestalt' (which we
found concerning).
●They were highly convicted in their sloppy views, and much of
their decision-making process for TTR-CM was inconsistent with
our understanding of the underlying biology and fundamentals.
As such, it's hard to extrapolate from their commentary or
predict precisely how the CARDIO-TTRansform results may
impact care broadly.
●While Attruby, in our view, likely offers the strongest event rate HR
(likely due to fastest monomer depletion) along with oral convenience,
BBIO is still battling intensely in a very split and competitive market
– which we expect will continue. We do give the company credit for
the execution thus far in such a challenging specialty, and expect this
momentum to continue.
●Given the size of the TTR-CM market, even a 4-way split means
multiple large blockbuster therapies – where everyone can be happy
