stored plasma from the ACS CPS-3 cohort. For 12 pre-specified cancer types that account

for approximately two-thirds of U.S. cancer deaths, the Galleri performed with 64%

sensitivity and 43% sensitivity within an average 1.36-year window prior to metastasis.

The authors conclude these estimates support MCED screening on an annual cadence

as a strategy to detect cancers before they reach late-stage disease.

NTRA investigators used tumor and matched-normal WES from 16,388 stage I–IV

CRC patients to map the germline and somatic molecular landscape across six genetic

ancestry groups (African, Native American, East Asian, European, Levantine Middle

Eastern, and South Asian). Ancestry was determined via "supervised local ancestry

inference" rather than self-reported ethnicity. MSI was the predominant hypermutation

mechanism (80.8% of hypermutated cases) and was more common among European-

ancestry patients. Significant ancestry-linked differences emerged in driver mutation

frequencies for APC, BRAF, KRAS, TP53, and PIK3CA in both MSI & MSS tumors.

Socioeconomic distress (measured by the Distressed Community Index) was higher

in African and Native American ancestry groups but did not independently influence

driver mutation frequency, suggesting the genomic differences are biologically rather than

socioeconomically driven.
