Why Do These Data Matter? Consistent Effects Regardless of Clinical

Presentation

●Taysha treated patients over the age of six, which is absolutely critical

as third party data/literature suggests that any treatment effects

observed at a younger age could be spontaneous, and we can’t

confidently attribute them to being drug related (the effect in TSHA’s

design to be spontaneous is between 0-6.7%).

●Today’s data show that regardless of age, clinical presentation, etc.

they are seeing consistent gains and improvements in developmental

milestones, skills, other improvements, etc.

●Another important conclusion is that the effects not only persist but

continue as time goes by. This demonstrates the MECP2 functions as an

“orchestra conductor” and that the expression of more genes become

regulated over time.

●We know Rett syndrome is a highly heterogenous disorder and the

effects TSHA is observing fall across different domains, and many

represent improvements that can change a patient and/or their

caregivers' lifestyle dramatically.

●This was also shown in two patient vignettes and several quotes/

examples shared by parents/caregivers (i.e., patient began walking

independently, reduced seizures, nonverbal to speaking in phrases and

multiple words, reduced stereotypies, using utensils, etc.).

●One thing that is leaving us very intrigued is the positive effects TSHA

is showing as it relates to seizure reductions. And while seizures

themselves are very problematic Taysha also tells us they can stun the

brain leading to difficulties in the ability to achieve new milestones.

●The company tells us to stay tuned for some deeper analysis on this
