ADA Sunday Takeaways: SYNCHRONIZEd or Asynchronous? Survodutide Data Leave Qs
Subgroup analysis from Regneron's COURAGE trial continued to highlight the potential lean
bodymass preservation benefits of both trevogrumab and garetosmab in patients both with and
without low lean mass at baseline. In today's analysis Regeneron broke down the COURAGE study
population by those with and without low lean mass at baseline with low lean mass characterized
by criteria derived from the FNIH sarcopenia project conference recommendations - more specifically
participants with appendicular lean mass/BMI of <0.789m2 in males and <0.512m2 in females. In their
analysis, Regeneron demonstrated that their treatment protocol enhanced lean mass preservation vs
semaglutide regardless of lean mass at baseline. While we find these data interesting, we continue to
look for more clarity on a potential path to approval for muscle preservation assets before we can begin
to give Regeneron more meaningful credit for the program.
Ph 1 monotherapy data shared today for Antag Therapeutic's AT-7687 (GIPR antagonist) bring
another novel agent into the fold that could help investors gain greater clarity on the agonism
vs antagonism debate. No weightloss curves were shared today because the presenter noted that no
weightloss was observed in these patients (inline with investigator expectations given the short length
of the study and the need to combine with other therapies to drive maximal weightloss). To underscore
this point the presentor highlighted some data the company had generated in NHPs where combination
of their GIPR antagonist with cagrilintide drove synergistic increases in weightloss - reaching weightloss
of 3.4%, 7.8% and 12.2% for '7687 mono, cagri mono and combo therapy respectively.
