Amylin's place in the obesity treatment paradigm still uncertain, but it's getting clearer. Net-net, we were

encouraged by KOL views of amylin as an exciting new tool in the toolbox. NOVOB's CagriSema will be the first

amylin to launch, with LLY's eloralintide on its heels.

A circulating hormone with a broad set of metabolic functions. Amylin is a peptide hormone co-secreted with insulin

from the beta cells in the pancreas that has long been used as an add-on to insulin for treatment of patients with diabetes

and advanced beta cell dysfunction. The receptor itself is a calcitonin receptor plus receptor activity modifying proteins

(i.e., RAMPS). While the gut hormones (e.g., GLP-1s) help you to feel satiated by slowing gut emptying to protect from

diarrhea and dehydration, amylin moderates blood glucose levels to prevent the over-release of insulin. And by going

to the brain (namely, the area postrema in the hindbrain), it enables the inhibition of appetite unique to the incretins on

the market today. Amylin tells your brain that you are satiated so you don't overeat, while also blocking glucagon so

you don't have dysregulation. Today, longer acting agents have been formulated as a potential mono- or combination

therapy in the setting of Type 2 diabetes, chronic weight management, and other comorbid conditions. Reach out to us
