ASCO26 "Under The Radar" Abstracts: Comp

Readthrough to WHWK, PRLD, STRO

We highlight key ASCO26 abstracts with comp readthrough to our coverage.

CPT113 linker/ payload ADC shows promising efficacy and safety, providing

validation for WHWK's ADC platform. In active KAT6 space, data from PFE

and OLMA strengthen case for PRLD's highly-selective KAT6a degrader as

potentially best-in-class. Comp TF and SEZ6 ADCs de-risk targets but leave

room for differentiated ADC approaches from STRO and WHWK.

WHWK

Validation of CPT113 ADC platform (Oral, Abstract #3000)

DXC006 (CD56 ADC with CPT113 topo1 payload) delivered encouraging Ph1 activity, with 69%

(n=36) ORR in SCLC, 46% (n=11) in lung adenocarcinoma, and 86% (n=14) in 2L SCLC. Safety also

looks promising vs. other topo1 ADCs, with low discontinuations (1.4%) and no ILD cases across

72 pts, which could be a meaningful differentiator if sustained. Despite Topo1 class-specific heme

tox with 21% Gr≥3 anemia, neutropenia notably did not appear in top-5 TEAEs (typically ~10-20%

for DXd ADCs). No ILD was observed

Overall, we view these data as strong validation of CPT113 ADC platform that WHWK has partnered on,

supporting the potency of this novel payload and hinting at improved therapeutic index pending longer

follow-up on ILDs, cytopenias, and durability. WHWK has 3 ADC programs using the CPT platform, with

HWK-007 (PTK7) and HWK016 (MUC16) expected to readout Ph1 in 1H27.

ABBV-706, SEZ6 ADC Comp in GBM (Poster, Abstract #2041)

ABBV-706 showed modest but meaningful activity in heavily pretreated GBM, with 8% ORR and 2

confirmed CR (4%), which is notable given poor prognosis of this population. Though WHWK is
